INTRODUCTION
Fc receptors are glycoproteins of approximate molecular weights of 50-70 kD. They are mostly present on white blood cells such as monocytes, tissue macrophages, B cells, granulocytes and some T cells. Fc receptors have great affinity for the Fc region of monomeric IgG (antibody) and non-specific staining and background staining can result from the binding of the Fc region of the antibody to the Fc receptors. Such examples of Fc receptor rich specimens are lymphoid sections, tonsil sections, bone marrow preparations and blood smears.
PRODUCT DESCRIPTION
Innovex Fc Receptor Blocker can be used as a blocking agent to block Fc receptors present on hematopoietic (blood derived) tissues and lymphoid tissues. The binding of the Fc region of the primary or the secondary antibody to the Fc receptors present on the surface of leukocytes (white blood cells) gives rise to non-specific staining in IHC and in flow cytometry assays. In addition, Fc Receptor Blocker can be used as a blocking agent for eliminating background staining in brain tissue and also tissues that have been high heat retrieved.
Innovex Fc receptor blocking agent is effective for blocking Fc receptors in lymphoid and hematopoietic tissues and smears that carry Fc receptors on their surface. Such specimen rich in Fc receptors are tonsil, bone marrow smears, lymph nodes and preparations made of blood components.
Fc Receptor Blocker is especially helpful for blocking Fc receptors in lymphoma cryostat and paraffin sections for CD markers staining and also for CD phenotyping in flow cytometric samples in leukemia specimens. Fc Receptor Blocker is also quite helpful for obtaining specific staining of IHC sections stained for kappa, lambda and Immunoglobulins (lgs).
APPLICATION/INTENDED USE
This reagent is intended to be used as a blocking agent for blocking Fc receptors in hematopoietic and lymphoid specimens and CD markers.

STORAGE CONDITIONS
Store in refrigerator at 2-8°C through the expiration date noted on the vial label.
INSTRUCTIONS FOR BLOCKING Fc RECEPTORS IN IMMUNOHISTOCHEMICAL AND IMMUNOFLUORESCENCE SPECIMENS:
- Deparaffinize slides or cut frozen sections, fix and rinse in water as usual.
- Quench endogenous peroxidase by immersion in 3% H2O2.
- Cover sections or smears with 3-6 drops of Fc receptor block to achieve full specimen coverage.
- Incubate for 30 minutes at room temperature.
- Rinse with rinse buffer.
- Proceed with immunostaining as usual.
INNOVEX Fc RECEPTOR BLOCKER CAN BE USED IN AUTOSTAINERS IN THE PRETREATMENT STEP.
INSTRUCTIONS FOR FLOW CYTOMETRY BLOCKING OF Fc RECEPTORS:
- Lyse or ficol blood as usual.
- Add 0.3 ml of Fc Receptor Blocker per 1,000,000 cells.
- Incubate for 20 minutes on ice.
- Wash twice in assay wash buffer.
- Proceed with antibody staining.
